Most banks charge overdraft for each item posted to your account that results in the balance being negative. And sometimes they will still cover the payment, and sometimes they return it as NSF (Non Sufficient Funds). If they do that, not only do you have the overdraft fee at your bank, but the store the payment gets returned to will want their money, and an additions fee ($25 to $35 usually). And you usually also have so many days to get the account back into the positive, or they will close your account and send what you owe to collections.
If this is your first time, ask your bank if they will waive the charge. They do understand that sometimes people make mistakes in balancing their account, so might be willing to help you by waiving the overdraft fee. Understand, though, that they are not required to, but you will never know unless you ask.
Usually banks will give you at about a day to realize you're in overdraft before charging you the fee. At my bank once I realized I was over I went in and asked about if they were going to charge me since pay day was a few days away. They said no, that there was a grace period of five days before they were going to charge me. I borrowed the money and covered the overdraft fee. If you contact your bank and ask them to waive it if it's the first time I would do that. People are more apt to work with others as long as the person reaches out.
I do not know their policy, but I've had a over draft fee from chase and they charged me 35 dollars. I called them and ask to waive the fee since it was the first time. They had no problem with doing that and said that they would usually allow it once a year. I did it again a few month later and they waived it for me again then told me the once a year thing again. I do not think they keep records of it, but I am more careful with my money now.
